Specific immunofluorescence staining of Treponema pallidum in smears and tissues.
To date, tissue sections prepared from Formalin-fixed tissues have not been successfully stained with Treponema pallidum subspecies-specific antibody in a direct fluorescent-antibody assay. While current methods stain T. pallidum, they do not distinguish T. pallidum from other spirochetes such as Borrelia burgdorferi (E. F. Hunter, P. W. Greer, B. L. Swisher, A. R. Simons, C. E. Farshy, J. A. C...
متن کاملImproved method for trypsin digestion of Paraplast sections before immunofluorescence staining.
Formalin-fixed, Paraplast-embedded tissue sections mounted on subbed slides were digested with crude trypsin in modified Sorensen phosphate buffer containing sodium fluoride before immunofluorescence staining. This method prevented detachment of sections from slides during processing, thereby permitting immunofluorescence and histological examinations of adjacent serial sections.
متن کاملVibration enhancement of slide-mounted immunofluorescence staining.
Immunofluorescence techniques allow the determination of protein and small molecule distribution within tissues and individual cells. There have been important, innovative modifications of these techniques since their introduction to the biosciences including the use of a mounting medium that prevents photo-bleaching, non-ionic detergents to permeabilize membranes, multiple immunofluorescence l...
متن کاملImmunofluorescence staining for detection of variola virus.
In September 1975 Bangladesh was the only country in the world with endemic variola major, and the eradication of the disease was imminent. A rapid and accurate laboratory diagnostic method was required to supplement immunodiffusion in agar gel and culture on chorioallantoic membrane of embryonated egg available at the Institute of Public Health in Dacca, Bangladesh. To determine its effectiven...
متن کاملIdentification of Rickettsia rickettsii in formalin-fixed, paraffin-embedded tissues by immunofluorescence.
With slight modification of a trypsin digestion technique, Rickettsia rickettsii were demonstrated specifically by immunofluorescence staining in Formalin-fixed, paraffin-embedded tissue sections from a human, rhesus monkey, and guinea pig with Rocky Mountain spotted fever and in infected membranes from a chicken embryo. Tissues were cut at 4 micron and, using geltain as a tissue adhesive, were...
